Large construct sequencing service

 

illumina

Full sequencing of your BAC, YAC or Cosmid clones by Shotgun Sequencing or Next Generation Sequencing is also available at Source BioScience LifeSciences.

 

Full sequencing of large constructs such as BAC, YAC or Cosmid clones/libraries can be challenging and time consuming using Sanger sequencing. Next Generation Sequencing using the , Illumina Hi Seq 2000, Illumina MiSeq & Roche 454 GS FLX+ System, has dramatically improved speed and reduced costs, enabling the full sequencing of large constructs at a fraction of the costs and time.

  • 1µg of extracted DNA

GLP-compliant sequencing

 

Requirements for the project

 

  • A study plan agreed between the customer and our Study Director
  • Template DNA, in the format of your choice, is supplied by the customer

 

The project itself:

 

  • A designated Study Director will oversee the comprehensive study plan
  • Specific primer design will be included within the study, if required
  • DNA preparation from a bacterial culture or agar plate, if necessary
  • Quality control of the template DNA
  • Sequencing will be performed on an ABI 3730 XL 96-capillary sequencer
  • Sequences will be analysed using "ABI base caller" which yields extremely accurate results
  • Depth of the final data, X-fold coverage, if required
  • All study and data material will be archived

 

On conclusion of the analysis:

 

  • A comprehensive report will be produced by our Study Director.
  • Chromatograms primer designs and any other electronic data can be downloaded from a secure ftp file and/or in CD format.

Shotgun sequencing service

 

Source BioScience LifeSciences can prepare libraries from samples including genomes, BACs, fosmids, cosmids and more. All genomic shotgun libraries are constructed and sequenced within our facility by our highly experienced scientists.

 

  • Large DNA fragments are nebulised and cloned into a standard E. coli vector
  • Only recombinant clones will grow due to the expression of the lethal gene, ccdB
  • Only circularised, single insert containing vectors will be propagated
  • T7, T3, M13f and M13r primer sites close to the insert site allows for more sequencing of the insert and not the vector

 

Source BioScience LifeSciences has completed large genomic sequencing projects on the mouse and pufferfish genomes and we are happy to provide our expertise to in sequencing and genomic assemblies.

 

  • Assembly using Staden GAP4
  • Sequencing will be performed on an ABI 3730 XL sequencer
  • Customer will receive the assembly and all reads in either disc, or ftp file format
